Schizophrenia remains a leading cause of global disability, typically manifesting in early adulthood and requiring lifelong clinical management [1]. While genome-wide association studies have identified numerous genetic variants linked to the disorder, translating these associations into actionable clinical targets has been hindered by the highly polygenic nature of psychiatric illness [2, 3]. Traditional research often relies on bulk brain tissue analysis, which averages out signals and may mask the distinct regulatory roles of individual cell populations like neurons and glia [4]. To cut through the noise of observational bias and reverse causation, researchers increasingly use Mendelian randomization, a statistical method that uses genetic variants as proxies for environmental exposures to determine causality [5, 6]. A recent study applies this causal framework to single-cell data, offering a highly granular view of how specific cellular pathways drive the development of schizophrenia and potentially opening new avenues for targeted pharmacotherapy.
