VTA-mPFC Dopamine Circuit Drives Stress-Induced Sleep Fragmentation in Mice

Sleep disturbances, particularly fragmented sleep, are a pervasive clinical challenge, frequently co-occurring with chronic stress and impacting patient quality of life across numerous medical conditions [1, 2]. Disrupted sleep is also a critical factor in the progression of neurodegenerative and neuropsychiatric disorders [3, 4, 5, 6]. While the link between stress and poor sleep is well-established [7, 8], the precise neural circuits involved have been unclear, limiting therapies to mostly symptomatic relief [9]. A recent study in mice provides a specific mechanistic explanation, identifying a key driver of this common clinical problem. The findings demonstrate that hyperexcitable dopaminergic neurons in the ventral tegmental area (VTA) lead to excessive dopamine release in the medial prefrontal cortex (mPFC), which in turn disrupts the stability of non-rapid eye movement sleep (NREMs) by degrading sleep spindles [10].
